Resumen: | [ES] Este trabajo se centra en la técnica de identificación de microorganismos MALDI-TOF,
enfocando su uso en la de levaduras enológicas y en concreto, comparando la población
microbiana de la vinificación tradicional con la de la maceración carbónica.
Desde hace años el método de vinificación clásico comienza por el despalillado y
estrujado de la uva; sin embargo, existe otra forma de elaboración, la llamada de maceración
carbónica. Es un proceso protagonizado por el metabolismo anaerobio que ocurre en las
bayas de uva intactas, sin estrujar, en ambientes enriquecidos en CO2.
La identificación de microorganismos presentes durante la vinificación permite tener un
mayor control de las elaboraciones y por lo tanto, asegurar un producto final de calidad. La
técnica de identificación microbiana MALDI-TOF se fundamenta en la espectrometría de
masas y se basa la separación de iones formados de acuerdo con su relación masa/carga.
El tiempo de vuelo de estos iones permite crear un espectro único basado en un perfil
protéico para cada microorganismo.
El hecho de que la técnica MALDI-TOF esté enfocada al entorno sanitario, nos ha
obligado a optimizar dicho equipo para su uso en levaduras enológicas. Por ello, el objetivo
de este estudio se ha centrado en comprobar la efectividad de la técnica para la identificación
de levaduras vínicas, complementándola con otra técnica identificativa ampliamente
utilizada, como es la secuenciación, y a continuación observar si existen diferencias
poblacionales entre la vinificación clásica y la vinificación por maceración carbónica.
Todo el proceso se ha llevado a cabo en el Instituto de las Ciencias de la Vid y el Vino
(ICVV) y el Complejo Científico Tecnológico de la Universidad de La Rioja durante la
Vendimia 2020. Gracias a este estudio, se ha concluido que MALDI-TOF es una técnica
adecuada para la identificación de levaduras enológicas, por la alta efectividad y fiabilidad
de los resultados, y por la rapidez con la que éstos se obtienen. Además, su precio por
muestra es menor al de otras técnicas. Cabe resaltar, que la clave de estos buenos
resultados es tener una base de datos completa y actualizada. [EN] This work focuses on the MALDI-TOF microorganism identification technique, focusing its use on the identification of enological yeasts more specifically, comparing the microbial population of traditional winemaking with carbonic maceration. For years, the classic winemaking method has started with the destemming and crushing of the grapes; however, there is another way of making it, the so called “carbonic maceration”. It is a process led by anaerobic metabolism that occurs in intact grape berries, without squeezing, in environments enriched in CO2. The identification of microorganisms present during the vinification process allows to have a greater control of the elaborations, and therefore, to ensure a final quality product. The MALDI-TOF microbial identification technique is based on mass spectrometry and is based on the separation of ions formed according to their mass / charge ratio. The time of flight of these ions allows to create a unique spectrum based on a protein profile for each microorganism. The fact that the MALDI-TOF technique is focused on the healthcare environment has forced us to optimize this equipment for use in enological yeasts. For this reason, the objective of this study has been focused on verifying the effectiveness of the technique for the identification of wine yeasts, complementing it with another widely-used identification technique such as sequencing, and observing if there are population differences between classic winemaking and winemaking vinification trough carbonic maceration processes. The entire process has been carried out at the Institute of Vine and Wine Sciences (ICVV) and the Technological Scientific Complex of the University of La Rioja during the 2020 Harvest. Thanks to this study, it has been concluded that MALDI-TOF is a suitable technique for the identification of enological yeasts due to their high effectiveness and reliability of results, the speed which they are obtained with, and their lower price per sample comparing it to other techniques. But it should be noted that the key to these successful results is having a complete and up-to-date database. |
