The Epidermal Growth Factor family in the ovary of European sea bass (Dicentrarchus labrax)

ResumenFollicle development and oocyte maturation are regulated by gonadotropins, ovarian steroids and growth factors produced by the ovary. The involvement of the epidermal growth factor (EGF) family in the local control of follicular growth and oocyte maturation is well documented in mammals, but less information exists in fish. In zebrafish, EGF family ligands are mostly expressed in the oocyte, whereas EGF receptors are exclusively expressed in follicular cells. In the European sea bass (Dicentrarchus labrax), a model marine perciform, the characterization of these paracrine factors has not been performed yet, and the functional relationship between endocrine and paracrine signaling systems still remains to be elucidated. Here we have studied the expression patterns of the EGF family ligands and EGF receptors (EGFRa and EGFRb) in different tissues of sea bass. Our results have revealed different expression patterns for each member of the family. All of them were detected in gonads, suggesting a role of this family in the control of gametogenesis in sea bass. Quantification of the expression of the EGF-like ligands and their receptors in the ovary of adult sea bass during a whole reproductive cycle showed different profiles for these genes. The expression of EGFRa, epigen (EPGN) and transforming growth factor alpha (TGFa) increases during autumn at the time of vitellogenesis progression, whereas the expression of EGFRb and betacellulin (BTC) is amplified at early winter, coinciding with the end of vitellogenesis. In all cases, except for EGF, maximum levels of expression are found in maturating-ovulating animals. Finally, the effect of FSH on the expression of the EGF family members in post-vitellogenic oocytes was studied. Supported by MICINN (AGL2008-02937, AGL2011-28890), GV (REPROBAS-Prometeo).
