Genome-wide nucleosome profile during mitosis and meiosis in fission yeast

AbstractThe eukaryotic genome is packaged in the nucleus as chromatin, whose fundamental units are nucleosomes. To study the role of nucleosome organization in the regulation of transcription, replication and recombination, we have generated genome-wide, highresolution maps of nucleosomes in Schizosaccharomyces pombe during mitosis and meiosis using tiling microarrays and next-generation sequencing. Our results show that most of the genome is occupied by positioned nucleosomes and that this pattern is largely conserved in mitosis and meiosis. We have identified approximately 3000 nucleosome-depleted regions (NDRs), of which 75% are identical in mitosis and meiosis. Global transcription analyses revealed local remodeling of the promoters of genes expressed at specific times during meiosis. We also found that high rates of transcription and antisense RNAs caused the delocalization of nucleosomes along the transcribed regions. Large intergenic regions (IGRs) often included clusters of nucleosome-depleted regions (NDRs), many of which colocalized with sites of double-strand breaks during meiosis. By contrast, the binding of the Origin Recognition Complex to A+T-rich replication origins did not colocalize with NDRs. Furthermore, poly (dA:dT) tracts displayed a tendency to map between nucleosomes but did not generate extensive NDRs. Together, our results show that the nucleosome organization of the S. pombe genome is to a large extent constitutive, and that very local remodeling takes place in response to the activation of the specific transcriptional programs during mitosis and meiosis.
