Abstract[EN] The present invention relates to a method for identifYing the tropism of the human immunodeficiency virus (HIV) in patients infected with said virus. The method includes joint culture of peripheral blood mononuclear cells (PBMCs) of a patient infected with HIV and PBMCs of a healthy control subject, in which the CD8-positive PBMC cells of the patient infected with HIV and/or of the healthy control subject are depleted. The virions released into the culture medium are inoculated by spinoculation to a human cell line isolated from the human body, which is CD4-positive and expresses the CCR5 co-receptor (CD4+/CCR5+/CXCR4-) and to a human cell line isolated from the human body, which is CD4-positive and expresses the CXCR4 co-receptor (CD4+/CCR5-/CXCR4+). Said method makes it possible to determine the viral tropism by quantifYing the proportion ofviral quasi-species that use one co-receptor or another.
[ES] La presente invención se refiere a un método de identificación del tropismo del virus de la inmunodeficiencia humana (VIH) en pacientes infectados con dicho virus. El método comprende el cocultivo de células aisladas mononucleares de sangre periférica (PBMCs) de un paciente infectado con el VIH y PBMCs de un sujeto control sano, donde se deplecionan las células PBMCs CD8 positivas del paciente infectado con VIH y/o del sujeto control sano. Los viriones liberados al medio de cultivo son inoculados mediante espinoculación a una linea celular humana aislada del cuerpo humano, CD4 positiva que expresan el correceptor CCR5 (CD4+/CCR5+/CXCR4-) ya una linea celular humana aislada del cuerpo humano, CD4 positiva que expresa el correceptor CXCR4 (CD4+/CCR5- /CXCR4+). Dicho método permite determinar el tropismo viral mediante la cuantificación de la proporción de cuasiespecies víricas que utilizan uno u otro correceptor.
