Physical map of bacteriophage ø29

Abstract

Restriction endonuclease EcoRI cleaves linear 29 DNA at four points yielding five fragments (A–E) of molecular weights 6.0 × 106, 3.8 × 106, 1.3 × 106, 0.6 × 106, and 0.3 × 106. The relative order of the fragments was shown to be A, B, E, D, C by analysis of the EcoRI partial digestion products and from the study of the fragments obtained by treatment of protein-containing DNA with EcoRI and trypsin. The EcoRI cleavage map of 29 DNA has been ordered relative to the genetic map by marker rescue experiments.
