Resumen: | [ES] La fase S del ciclo celular, es la fase donde el ADN se replica. Los errores en esta fase alteran
la viabilidad celular y producen inestabilidad genómica. Además, en esta fase existe un punto
de control: el checkpoint de fase S, regulado en Saccharomyces cerevisiae por las quinasas
Mec1 y Rad53 desencadenando la respuesta del checkpoint. La replicación del ADN se realiza
por tres ADN polimerasas: Polα inicia la síntesis y es sustituida por Polε en la replicación de la
cadena continua y Polδ en la cadena discontinua. Las polimerasas pueden introducir
ribonucleótidos (rNTPs) por error siendo eliminados por la subunidad catalítica de la
ribonucleasa RNasa H2. En situaciones de estrés en la replicación, se activa el Checkpoint de
fase S llevando a cabo una respuesta coordinada. Cuando el Checkpoint no funciona
correctamente Exo1 es responsable del colapso de las horquillas de replicación.
El objetivo de este estudio era ver si Exo1 afectaba de manera diferencial a la estabilidad de
las cadenas continua o discontinua cuando hay estrés replicativo. También se quiso estudiar el
papel de la fosforilación de Exo1, realizada por Rad53, en las cadenas de nueva síntesis.
Para ello, existen técnicas como Pu-seq que permiten analizar la síntesis de las cadenas
nacientes por las distintas polimerasas. Por ello, es necesario construir cepas que tengan
mutaciones en las DNA polimerasas Polδ y Polε, para promover la introducción de mayores
niveles de ribonucleótidos durante la replicación del DNA, así como mutaciones en Rnh201
que eliminen la ruta RER, y por tanto las células no puedan eliminar el exceso de rNTPs. La
técnica Pu-seq se basa en las roturas del DNA a nivel de rNTPs por hidrolisis alcalina, y el
posterior análisis por secuenciación de los fragmentos generados a nivel genómico. [EN] The S phase of the cell cycle is the phase where the DNA replicates. Errors in this phase alter cell viability and produce genomic instability. In addition, there is a checkpoint in this phase: the S-phase checkpoint, regulated in Saccharomyces cerevisiae by the Mec1 and Rad53 kinases, triggering the checkpoint response. DNA replication is performed by three DNA polymerases: Polα initiates the synthesis and is replaced by Polε in the continuous chain replication and Polδ in the discontinuous chain. Polymerases can introduce ribonucleotides (rNTPs) by mistake and are removed by the catalytic subunit of ribonuclease RNase H2. In situations of replication stress, the S-phase Checkpoint is activated, carrying out a coordinated response. The objective of this study was to see if Exo1 differentially affects the stability of the continuous or discontinuous chains when there is replicative stress. We also wanted to study the role of Exo1 phosphorylation, performed by Rad53, in the new synthesis chains. For this purpose, there are techniques such as Pu-seq that allow to analyze the synthesis of the nascent chains by the different polymerases. Therefore, it is necessary to build strains that have mutations in the DNA polymerases Polδ and Polε, to promote the introduction of higher levels of ribonucleotides during DNA replication, as well as mutations in Rnh201 that eliminate the RER pathway, and therefore the cells cannot eliminate the excess rNTPs. The Pu-seq technique is based on DNA breaks at the rNTP level by alkaline hydrolysis, and the subsequent analysis by sequencing of the fragments generated at the genomic level |
