Characterization of culturable marine luminescence bacteria isolated from Gulf of Nicoya, Costa Rica

AbstractBacterial luminescence in bacteria is dependent on quorum sensing and is regulated by the luxCDAB operon. The expression of this operon genes can be inhibited by exposition to toxic substances such as heavy metals. Therefore, luminous bacteria have been used in aquatic ecosystem toxicity tests. In this study we determined the abundance and identity of luminescent bacteria from coastal waters of the Gulf of Nicoya (Costa Rica). We also evaluated the preliminary effect of heavy metals on luminescence intensity. Samples were collected from four sites of the gulf, filtered and cultured in marine agar. Total culturable heterotrophic and luminescent bacteria were determined by enumerating total colonies growing in daylight and dark conditions respectively. The luminescent strains were subjected to phenotypic and phylogenetic characterization, which included biochemical tests and multilocus sequences analysis using 16S rRNA, gyrB and pyrH genes. The effect of heavy metals on growth and luminescence was determined by the disk diffusion test. Culturable luminescent bacterial ranged from 0-10% of total heterotrophic bacteria and no significant differences between sites could be observed. The isolates belonged to the genera Photobacterium and Vibrio. We identified in toxicity tests, various strains with high luminescence and defined inhibition zone that showed potential as indicators for monitoring environmental toxicity
