Water soluble carbohydrate profiling in oat (Avena sativa) point to fructans and raffinose family as key metabolites for drought tolerance

ResumenCarbohydrates are of essential importance in plant energy metabolism as well as structural polymers. Furthermore, recently they have been also proposed to play an important role in tolerance to biotic and abiotic stresses. In a previous metabolomic work (Sánchez-Martín et al. 2015), we observed an overall differential response in several carbohydrate and energy metabolism pathways between drought tolerant and susceptible oat genotypes. In the present work, during a time course of increasing water deficit, carbohydrates from leaf samples were profiled using gas chromatography-mass spectrometry (GC-MS). Carbohydrate profiles of the tolerant genotype highlighted higher accumulation of monosaccharides (fructose and glucose) and also oligosaccharides (fructans and raffinose), compared with the susceptible genotype following water deficit. No differences were found for mannitol or trehalose, sugars previously reported to have a putative role during stress responses. Both, fructans and raffinose family, contrary to starch, are osmotically flexible as their polymerization degree may easily be changed. This may facilitate lowering osmotic potential, contribute to water inflow and help to stabilize cell membranes. Further work to test this hypothesis in oat is in progress.
