The Exportin5-aatRNA-EF1A complex mediates the nuclear export of SNAG-containing proteins

AbstractExportin 5 mediates the nuclear export of dsRNAs including pre-miRNAs, adenoviral RNAs and tRNAs. When tRNAs are aminoacylated, the aatRNA-Exportin5 complex recruits and co-exports the elongation factor eEF1A. Through the combination of nuclear export assays and yeast double-interaction screens, we show that eEF1A recruits Snail transcription factors in the nucleus, thereby being co-transported by the aatRNA-Exportin5 complex to the cytoplasm. Snail binds to eEF1A through the SNAG domain, a novel protein nuclear export signal (NES). This new complex provides a mechanism to remove SNAGcontaining transcription factors from their target promoters limiting their activity and exporting them to the cytoplasm.
