Direct method for the analysis of Cysteinated and Glutationated precursors of 3 MH and 4MMP by UHPLC-MS

AbstractDuring vinification and aging ofwine a large number of aromas are generated, some of which come from odorless precursors present in grapes. So me of these precursors are molecules bound to cysteine and glutathione. They are !he precursors of varielal polyfunctional mercaptans: 4-mercapto-4-methyl-2-pentanone, 4-mercapto-4- methyl-2-pentanol and 3-mercaptohexanol. These precursors have low leve! concentrations in nmsts, but they potentially have a great sensory importance, due to the low olfaction threshold of the molecules released from them. [ 1] To date, all methods applied to determine cysteine and glutathione precursors relied on extraction methods like SPE. The method proposed in this paper allows direct analysis of these precursors in must, without any previous isolation step, by using ultrahigh resolution liquid chromatography coupled with mass spectromel!y (UPLCMS). The method quantifies in 20 minutes the 4 precursors with a simple filtration of the musts. The LQ for 4-mercapto-4-methyl-2-pentanone-L-cysteine is between 0.6 and 3 g 1 L, 3-mercapto-hexan-1-ol-L-cysteine between 0.3 and 1.75 mg 1 L, 4-mercapto- 4-methyl-2-pentanone-glutathionebetween 0.3 and 0.6 g 1 L between 1 and 3- mercapto-hexan-1-ol-L-glutathione, 2 and 7 mg 1 L. Reproducibility (expressed as RSD) of the method for analysis of grape musts is under 13% in all cases except for mercapto-hexan-1-ol-L-glutathi one. The proposed analysis method was employed to study the influence that different treatments perfonned in the vineyard have in the synthesis of glutathione and cysteine precursors. The grape varieties Chardonnay and Gewurztraminer have been studied. For variety Gewurztraminer cysteine derivatives present h.igher concentrations in defoliated grapes, whereas the mercapto-hexan-1-ol-L-glutathione presents higher concentrations in witness grapes. For Chardonnay variety results are notas clear, as the effect is different for the three precursors quantified.
