ResumenPelagophytes are important components of marine photosynthetic picoeukaryote assemblages, including members widely distributed across the oceans. Molecular surveys based on ribosomal genes suggest that pelagophytes have a low evolutionary diversification, with sequences typically close to cultured species, mostly Pelagomonas calceolata. To assess the intraspecific variability of this group we analyzed pelagophyte specific pyrosequencing reads containing a part of the 18S and the complete ITS1 region, the later used as a higher resolution phylogenetic marker. Our sampling includes open ocean sites of major basins and coastal sites around Europe. Most pelagophyte genera were represented within our dataset and a novel group without cultured representatives was detected. The majority of OTUs were related to Pelagomonas calceolata and to Aureococcus anophagefferens, accounting together ~80% of the reads. Pelagomonas was present and dominant in most of the sampling sites, which confirms the broad distribution of this species in marine systems. Aureococcus was more restricted to coastal sites. Preliminary analysis of the ITS1 region of P. calceolata indicates low intraspecific variability and suggests that this species has a world-wide population without ecotypic differentiation
