Topoisomerase II and condensin activate a DNA damage independent mitotic checkpoint in chromatin assembly mutants

ResumenThe maintenance of the chromatin structure is essential for genomic integrity and cell cycle progression. Consequently, a reduction in the levels of histones causes defects on chromatin assembly that are associated with fork instability, DNA damage, mitotic arrest and loss of viability. This mitotic arrest is mediated by the non-overlapping activities of Mec1, Tel1 (the major sensors of the replicative and DNA damage checkpoints) and Mad2 (an essential component of the spindle assembly checkpoint; SAC), suggesting that the attachment of the chromosomes to the spindle is also affected by histone depletion. However, checkpoint activation in these cells is not associated with Mad2-GFP foci at centromeres, a key feature of SAC. Strikingly, a reduction in the activity of either topoisomerase II or condensin, involved in the maintenance of chromosome structure during genome condensation, partially suppresses the mitotic arrest and loss of viability but not the accumulation of DNA damage associated with histone depletion. These results support the existence of a topological checkpoint that detects condensation problems arising as a consequence of defective chromatin assembly.
