Transcriptomics and metabolomics of wild and resistant human leukemia cell proliferation. A Foodomics evaluation of dietary polyphenols effect

ResumenIn this work. a Foodomics strategy is applied to investigate the anti-proliferative effect of dietary polyphenols from rosemary on two human leukemia lines. one showing a drugsensitive phenotype. and another exhibiting a drug-resistant phenotype. Namely, whole-transcriptome microarray together with a MS-based non-targeted analytical approach (via capillary eiectrophoresis time of flight mass spectrometry. CE-TOF MS, and ultrahigh performance liquid chromatography-time of flight mass spectrometry, UPLC-TOF MS) were employed to carry out transcriptomics and metabolomics analyses. respectively. Functional enrichment analysis was done using ngenuity Pathway Analysis (PA) software as a previous step for a reliable interpretation of transcriptomic and metabolomic profiles. Integration of data obtained from transcriptomics and metabolomics platforms was done by overlaying datasets on canonical (defined) metabolic pathways using PA software. This strategy enabled the identification of several differentially expressed genes alone andlor linked to the changed metabolic pathways that were modulated by rosemary polyphenols providing new evidences at molecular leve1 on the effect of these compounds.
