AbstractThe aldol addition reaction has long been recognized as one of the most useful tools that has the synthetic chemist for the construction of new carbon-carbon bonds. The enzyme aldolases are among the most important biocatalysts for C–C bond formations in nature. Dihydroxyacetone phosphate-dependent aldolases have been extensively used in chemoenzymatic syntheses of carbohydrate, carbohydrate-like structures, or non-carbohydrate compounds. This review will be mainly focused on that applications in which DHAP-dependent aldolase are part of a multi-enzyme system, or alternatively on those in which the aldolase catalysed reaction is key in a multi-step synthetic pathway.
