Resumen: | Parkinson’s disease (PD) is one of the most common neurodegenerative disorders characterized by the loss of dopaminergic neurons in the substancia nigra pars compacta. The molecular mechanisms underlying the pathogenesis of PD are complex and still unknown, but it has been suggested that mitochondrial dysfunction is an important feature of the disease. It is still debated whether the observed dysfunction is involved in the onset and progression of the disease or if it is a consequence of neurodegeneration. Here, we evaluate mitochondrial morphology and mitophagy in a PD genetic model in order to determine if differences exist depending whether patients manifest or not PD. We have found that mitophagy levels are decreased in skin fibroblasts from PD manifesting-carriers in comparison to non-manifesting carriers. Moreover, our results showed that mitochondrial networks are more fragmented in fibroblasts from patients that manifest PD, as the mean number of branches per network is decreased. In contrast, we have not demonstrated differences in mitochondrial footprint. Altogether, our results suggest structural and morphological mitochondrial differences, without affecting the total mitochondrial amount, in fibroblasts from carriers of LRRK2G2019S mutation and manifesting PD when compared to non-manifesting individuals | URI: | http://hdl.handle.net/10261/177869 |
