Fine-scale phylogeographic analysis of the occidental clade of the Pyrenean desman

AbstractThe Pyrenean desman ( Galemys pyrenaicus ) is a small semi-aquatic mammal endemic to the northern part of the Iberian Peninsula. It inhabits clean rivers and streams and is endangered in part of its distribution range. Previous studies showed a strong genetic structure, with four mitochondrial clades that revealed a very small degree of mixing. Here, we studied the occidental clade, which is the most diverse, with the objective of performing more detailed analyses about the mitochondrial genetic diversity and past dispersal patterns of this species. To do so, we used more than 200 samples, mostly non-invasive, either from desman faeces or from otter ( Lutra lutra ) faeces containing depredated desmans. We sequenced cytochrome b and D-loop fragments and performed different phylogenetic and phylogeographic analyses. With this larger dataset, we described better the type of mixing that takes place in the contact zone of the two major mitochondrial clades in the Cantabrian Mountains. In addition, the more detailed map of genetic variability allowed us to detect areas with lower diversity, which may have been more recently colonized. Another objective of this study was to quantify the degree at which river basins are responsible for the genetic differentiation at this scale. All this information could help to improve the conservation status of this rare endemic mammal.
