dc.description.abstract | [Purpose]: Tumor-derived exosomes are a significant mechanism of intercellular communication, affecting tumor-related pathways, such as angiogenesis, drug resistance and immune response. This study analyzed the release of exosomes from breast tumor cells with different capacities of stemness/metastasis based on CXCR4 expression and evaluated their involvement in the capacity of generate oncogenic features in recipient cells. [Experimental design]: Oncogenic effects of breast cancer cells-derived exosomes were evaluated in vitro and in vivo models. Exosome cargo in mRNAs involved in stem cell differentiation and metastasis were analyzed by specific quantitative PCR arrays. Finally, stemness- and metastatic-related mRNA most differentially detected in exosomes were analyzed in plasma of breast cancer patients assessing its capacity as a prognosis marker. [Results]: In vitro, exosomes released from CXCR4-tumor cells modify stemness markers, and proliferation, migration and invasion capacities of neighboring cells. Accordingly, inoculation of CXCR4-cells-derived exosomes in immunocompromised mice mediated primary tumor growth and metastatic potential. Exosomes isolated from CXCR4-cells and stemlike cells were highly enriched in stemness- and metastatic-related mRNAs and showed high homology between them. Furthermore, comparative analysis of mRNAs contained in exosomes isolated from patients revealed a gene signature of mRNAs highly enriched in exosomes of patients with worse prognosis. [Conclusions]: Our data support the view that exosomes released by CXCR4-cancer cells that exhibit stem-like properties are capable to transfer these oncogenic features to recipient cells. In addition, our results show the involvement of >stemness and metastatic signature> mRNA in exosomes from plasma in breast cancer patient survival. | - |
