Dual targeting to mitochondria and plastids of AtBT1 and ZmBT1, two members of the mitochondrial carrier family

AbstractZmBT1 and AtBT1 are members of the mitochondrial carrier family (MCF) that are suggested to be exclusively localized in the envelope membranes of plastids. ZmBT1 is thought to be involved in the transport to amyloplast of cytosolic ADPG linked to starch biosynthesis in maize endosperms, whereas AtBT1 appears to be involved in the export from plastids to the cytosol of newly synthesized adenylates in Arabidopsis. To investigate the impact of ZmBT1 and AtBT1 in plant metabolism, in this work we produced potato, barley and Arabidopsis plants ectopically expressing Zmbt1 and Atbt1. Zmbt1--expressing plants produced variegated leaves with reduced starch content, whereas Atbt1-expressing plants produced normal leaves accumulating wild type starch content. Homozygous Atbt1::T-DNA mutants displayed aberrant growth and sterility phenotype, which was complemented by Atbt1, but not by Zmbt1. The overall data thus indicated that (a) Zmbt1 and Atbt1 have different functions and (b) Atbt1 is an essential gene in Arabidopsis. Confocal fluorescence microscopy analyses of potato, Arabidopsis and maize plants stably expressing green fluorescent protein (GFP) fusions of ZmBT1 and AtBT1 revealed that the two proteins have dual localization to plastids and mitochondria. Furthermore, analyses of plants expressing GFP fusions of the ZmBT1 and AtBT1 N-terminal extensions, and of the mature proteins revealed that (a) AtBT1 and ZmBT1 N-terminal extensions comprise targeting sequences exclusively recognized by the plastidial compartment, and (b) targeting sequences to mitochondria are localized within the mature part of the protein. To the best of our knowledge this is the first report showing dual localization of MCF proteins to plastids and mitochondria. Based on these results, we propose that functions of ZmBT1 and AtBT1 should be re-evaluated.
